چکیده

Metallothionein (MT) has a dynamic role in cellular zinc metabolism. In order to determine the apoprotein thionein (T), an assay was developed. With this method tissue homogenates were treated with subtilisin to digest any T that might exist but MT could not be digested by subtilisin. MT was detected by cadmium-hemoglobin saturation method and T was determined as the difference of MT between samples with and without subtilisin added. With this assay the value of T/MT was found as 0.67, 0.53, 0.14 and 0.10 in four different hamster tissues brain, testis, liver and kidney, individually. This finding was confirmed by gel filtration experiments of liver homogenate supernatants. Upon addition of zinc to the homogenate, zinc in MT containing fractions increased about 2.5 fold. Since the tenet has been that the steady state concentration of T in cells is very low due to its rapid proteolytic digestion, the existence of T under normal physiological condition is a novel finding that has important implications for the function of this protein in zinc metabolism. Cd2+-induced synthesis of metallothionein in HeLa cells.

HeLa cells synthesize metallothioneins in response to Cd2+. The kinetics of thionein (apoprotein of metallothionein) synthesis was studied by pulse-labelling the cells with [35S]cysteine and measuring relative amounts of the labelled thioneins separated by electrophoresis. Thionein synthesis rapidly increases in the first 6-8 h after exposure to 0.5 micrograms of Cd2+/ml, and begins to decrease...

Metallothionein synthesis and degradation: relationship to cadmium metabolism.

Metallothionein is an integral component of the mechanism that regulates the metabolism of cadmium and zinc. The synthesis of this protein can be "induced" by oral or parenteral administration of either metal. The metallothionein mRNA content of liver polysomes is increased shortly after an influx of small amounts of either metal into hepatocytes. After sufficient amounts of this poly (A+) RNA ...

Cell-free synthesis of metallothionein directed by rat liver polyadenylated messenger ribonucleic acid.

Total RNA was isolated from rat liver polyribosomes and fractionated by oligo(dT)-cellulose chromatography to obtain polyadenylated mRNA. The mRNA was translated in a wheat-germ cell-free protein-synthesizing system containing [3H]glycine, [3H]lysine and [3H]serine. Most of the newly synthesized 3H-labelled polypeptides were removed from the cell-free products by precipitation at pH 4.0. 3H-lab...

Cellular zinc and redox states converge in the metallothionein/thionein pair.

The paramount importance of zinc for a wide range of biological functions is based on its occurrence in thousands of known zinc proteins. To regulate the availability of zinc dynamically, eukaryotes have compartmentalized zinc and the metallothionein/thionein pair, which controls the pico- to nanomolar concentrations of metabolically active cellular zinc. Interactions of zinc with sulfur ligand...

The Zn- or Cu-Thionein Character of a Metallothionein Determines Its Metal Load When Synthesized in Physiological (Metal-Unsupplemented) Conditions

The present work comprises the recombinant synthesis of four metallothioneins (MTs) in metal-unsupplemented cultures and the characterization of the recovered metal complexes by means of analytical and spectrometric techniques. The four MTs are two Drosophila (MtnA and MtnB), one yeast (Crs5), and one mouse (mMT1) metallothionein isoforms. These four MTs exhibit distinct metal binding preferenc...
